Für das Qualitätskontroll (QK) -Labor des "Centrum für innovative Therapie" (CIT), soll ein hochauflösendes Massenspektrometer gekoppelt an ein UHPLC-System (LC-HRMS) zur Reinheit-, Identitäts- und Gehaltsbestimmung von Peptid-Vakzinen angeschafft werden.

Lots Lot 1 — LC-MS System: Die wichtigsten Parameter, die bei der In-Prozess-Kontrolle und zur Freigabe der hergestellten Wirkstoffe und Prüfpräparate (Investigational Medicinal Products (IMPs) - Arzneimittel für klinische Studien) auf dem LC-HRMS Gerät ermittelt werden sollen, sind die eindeutige Bestimmung der Identität mittels der exakten Masse und Isotopenverteilung, Bestätigung der Peptidsequenz durch MS/MS Experimente, Detektion, Identifikation und Quantifizierung von z.T. isobaren Nebenprodukten, sowie die Gehaltsbestimmung des Ziel-Peptids. Aufgrund der großen Zahl potenzieller Analyten in den Zwischen- und Endprodukten, bei u.U. sehr schneller Elution (UHPLC), ist die Geschwindigkeit des Geräts bei gleichzeitig hoher Auflösung entscheidend. Quadrupol/ Time-of-Flight (QTOF) - Geräte kombinieren beide Aspekte und stellen somit den für unsere Zwecke geeignetsten Massendetektor dar.
